clone existing WinToUSB as backup

I am aware of the procedure to clone an existing Win 10 USB to another disk as backup, but have a question.  I have enabled the hard disk of the host computer in my existing USB.  Should this be disabled before creating a new clone or might the software do this automatically?

Don't worry about it, this will not have any effect on cloning Windows.
